What really jumps out here is framing genetic tool development as a search problem instead of a design problem. Most labs treat non-model organism work like they need to rationally engineer the perfect plasmid, but thats slow and often fails when expression contexts dont transfer. The barcode pooling approach sidesteps that entriely by letting empirical data reveal what actually works across the variants. Ive been in situations where we spent months optimizing a single construct for a novel host, wish we'd had something like this.
